Fibrinogen is a vital biopharmaceutical, but purification has remained problematic. GPRPK pentapeptide is a synthetic ligand designed to bind fibrinogen, it can be immobilised on Fractogel and used over 50 times without losing capacity and achieving over 95% protein purity. Fibrin fragments can also be purified from plasma due to the recognition of the GPR residues. This has been clinically applied to help atherosclerosis patients by removing fibrinogen from the plasma, reducing the viscosity, and thus improving microcirculation blood flow. A batchwise process of fibrinogen production has been superseded in efficiency by the generation of GPRPK bound to a novel sucrose-based polymer (SUBPOL) for affinity chromatography.
