CIGB-300 is an anti-casein kinase 2 (CK2) peptide. CK2 is a serine-threonine protein kinase with more than 300 substrates which plays a role in several cellular processes including cell growth and proliferation, cell viability, and apoptosis. CK2 is also frequently dysregulated in many human tumours phosphorylation by CK2 is a biochemical event involved in human oncogenesis. CIGB-300 binds to the phospho-acceptor domain of CK2 substrates, impairing the correct phosphorylation by the enzyme, thus reducing tumour cell adhesion, migration and invasion. CIGB-300 therefore has potential as a cancer therapy. P15 is fused to the cell-penetrating peptide derived from the HIV-Tat protein that has been extensively used for delivering peptides cargoes within cells. P15-Tat induces apoptosis in a variety of tumour cell lines.
