Collagen-mimetic signal peptide for structural ECM remodeling

Hexapeptide-9 (Collaxyl®) is a collagen-inspired hexapeptide with a Gly-Pro-Gln repeat motif that mimics the structural collagen triple-helix sequence found naturally in human Collagen IV and XVII. It stimulates fibroblast proliferation, activates COL1A1 and COL3A1 gene expression, and modulates integrin expression to enhance fibroblast-ECM adhesion. A landmark 2025 randomized, double-blind clinical trial demonstrated that cyclized Hexapeptide-9 significantly outperformed retinol in reducing the number, area, and roughness of both crow's feet and forehead wrinkles.

Hexapeptide-9 works through a dual mechanism: direct gene activation of structural collagen genes and improved fibroblast attachment to the ECM via integrin modulation. This results in a more robust and dynamic skin structure. The 2025 clinical trial (DOI: 10.1111/jocd.70290) represents one of the most rigorous clinical confirmations of peptide efficacy, comparing directly against retinol in a controlled, blinded design. Produced via SPPS to ≥95% purity (HPLC + MS). Stable at pH 4.5–7.5.
