Palmitoyl Pentapeptide-4 (Matrixyl®) is the world's most extensively studied and commercially successful cosmetic signal peptide, introduced by Sederma in 2000. The KTTKS sequence is a naturally occurring procollagen I C-propeptide fragment (matrikine) that signals fibroblasts to synthesize Collagen I, III, and Fibronectin. The palmitoyl group ensures stratum corneum penetration. A rigorous 12-week split-face randomized controlled trial demonstrated significant improvements in wrinkle depth and skin roughness versus vehicle control.

Main characteristics

Palmitoyl Pentapeptide-4's matrikine mechanism mirrors the body's natural collagen synthesis feedback loop: when collagen is processed, the released KTTKS fragment signals fibroblasts to produce more. Matrixyl mimics this endogenous signal exogenously. Multiple independent human studies confirm consistent anti-aging efficacy without the irritation associated with retinol.
