Pentapeptide-18 (Leuphasyl®) is a synthetic enkephalin-mimetic pentapeptide that helps reduce the appearance of expression wrinkles through a mechanism distinct from botulinum toxin-like peptides. Structurally related to the opioid peptide Dalargin, it interacts with opioid receptors linked to inhibitory G proteins (Gi), modulating neuronal signaling and reducing neurotransmitter release. By influencing calcium-dependent pathways involved in muscle contraction, Leuphasyl® helps soften dynamic wrinkles and is often combined with other neuromodulating peptides in anti-aging formulations.

Main characteristics

Pentapeptide-18's mechanism is complementary and distinct from Argireline: while Argireline blocks SNARE complex assembly (pre-synaptic), Leuphasyl activates opioid receptors to reduce acetylcholine release via G-protein-mediated calcium channel inhibition (a different pre-synaptic mechanism). This mechanistic complementarity justifies their combination in anti-wrinkle formulations. Stable at pH 4.0–7.5. Produced via SPPS with D-Ala modification to ≥95% purity (HPLC + MS).
